Well it can work together since the scan engine of Defender is limited to

some extent ( regular malware ) in which mcafee can compensate ( to deal with

some more complexed malicious files like PE infectors and others ) also

Mcafee has a separate engine.

 

The only rule of the thumb no 2 antivirus is advised to run because it can

mess up the process capability of the system
