Help! I cannot access a file that I need!

I need to access C:\Documents and Settings\[My Username Here]\Local Settings\Application Data\Microsoft but when when I click on [My Username Here] in Documents and Settings, it comes up with this error message. C:\Documents and Settings\[My Username Here] is not accessible. Access is denied. How can I fix this?
